What parameters does a NGS-Sequencing run have?

Hey guys,

i am currently investigating using NGS-Sequencing in my project.

What parameters/conditions do i need to know about that could influence my result?

Parameters i know about so far:

- number of cycles
- temperature
- MgCl, DMSO- Concentration
- Polymerase

A few more things to add to your list:

• Type of sequencer
• Reagents used for library prep.
• Person doing library prep.
• Single/paired-end
• Quality of samples

That's a non-exhaustive list.
