Details, please! Which organism? Is this one gene or two? Do you want to carry the clone in bacteria or another organism? Provide a link to the gene(s) or tell us how long, how many exons they are.

I suppose the part of the question on how to design/choose the primer sequences could be considered a bioinformatic problem. You could re-formulate your question to ask what bioinformatic tools are available to help you design good primers for your gene of interest.
