Question: Best way to determine if two groups of sequences are homologs?

Hello!

I had a question - what program/method would you use to determine if two groups of sequences (for example: group a with 7 sequences and group b with 6 sequences) are homologs? Would you use BLAST or ClustalOmega alignment? I think the thing is, I'm confused about which program is best and how to determine homology from that program.

Additionally - if not a bioinformatics program - what can you look for to determine homology?
