How is an RNA template to be reverse transcribed into cDNA chosen?

I read the paper, "Bioluminometric Method for Real-Time Detection of Reverse Transcriptase Activity", and was wondering how they chose the E2PN template oligonucleotide? I'm assuming it was partially chosen to not match any known genetic sequences. I'm not sure of the design process in general though?
