I don't think there is a tool which satisfies your three requirements.

If you are really interested in bioinformatics then you MUST start using UNIX based OS, most tools nowadays are developed to be run in command line (as it can be integrated into a pipeline easily). Few options for windows OS would be:

  1. Install virtual machine and then download Biolinux and use it with virtual box.
  2. Use Galaxy web platform.

There are other paid options like CLC Genomics workbench. But as I mentioned earlier, the options for windows based users are very limited.
