Dear Sir/Madam, I have calculated the interaction energy between protein and another protein in a protein-protein complex using foldX. The value obtained is -0.13 and in another case I obtained the value of interaction energy as -8.
Interaction energy as per calculated by foldX ∆∆Gab = ∆Gab- (∆Ga + ∆Gb)
If I want to predict the stability from interaction energy by foldX, which value of interaction energy should I consider as stable. If the interaction energy decreases from -0.13 to -8, can I say that the binding affinity increases? It would be great if you please cite some references where I can use interaction energy to obtain the desired protein stability.
