I want to classify single copy genes and duplicate genes in whole Human genome . But I do not find a good way to solve this problem.

Rreciprocal best blast hits is one good way to do this. Blast each gene against all genes and compare the results of the best hit. See the work done in Koonin's group in the early 2000s (e.g. https://www.ncbi.nlm.nih.gov/pubmed/12969510) or the EggNOG database http://eggnogdb.embl.de/#/app/home
