with what technique could I know if an RNA is non-coding?

How could you know the size of a genome before amplifying it?

We all know that there are many programs to study the assembly of genes, but which are really good criteria to check it?

You want to know which genes are coding and non-coding in an annotated or unannotated genome?

You can estimate genome size with kmer profiles (see a tutorial here and here, or search the web for more details), or using flow cytometry. Of course, genome size estimation with kmer profiles depends on generating sequencing data.
