Hello, I was asked to perform dissociation curve analysis with SYBR Green on Rotorgene instrument. I've never done this, so I do not understand, where to start. First, I have to do qPCR reaction with pcr mix, gDNA and primers. And what after? Leave the plate in cycler and perform steps like these? Step Ramp rate Temperature Time 1 1.6°C/second 95°C 15 seconds 2 1.6°C/second 60°C 1 minute 3[1] 0.15°C/second 95°C 15 seconds
Can I set this step in one run with PCR reaction?
