can anybody give a clarification on the fact that we need to do duplicates or triplicates for lncRNA sequencing? single set of sample is enough or not?

I suggest you google on the basics of experimental design. Unreplicated experiments are never sufficient to make any claims no matter if you do a low-throughput assay or NGS. Replicates are always necessary to assure robustness and reproducibility of your results. Be sure to get a solid background in experimental design first and try to talk to experience people before conducting any experiments.
