Hello,

I am wondering if someone could guide me or recommend me some reading material to understand what sort of enrichment (if any) can I expect from a paired-end Illumina TruSeq RNAseq sequencing. I mean, is there a 3' or 5' enrichment that I should be aware of? (as for example 10x does in their 5' or 3' library prep kits).

Also, I would love to start a discussion on how to test this -- For future reference and for different library preparation kits.

Thanks!

RNAseq blog has several articles related to bias (LINK).

Are you trying to understand the different library preparation methods that are typically used for bulk RNA-seq? This review paper by Griffith et al (2015) is a great place to get started.
