How to calculate Minion run time needed?

Hi all,

Does someone know how to calculate how many hours (approximately) i need to run an minion, to get a good coverage for example of bacterial genome with 3M bases of size? We want to do two separate runs with wash, in order to sequence 2 bacterial genomes, so i need to know how much time to run the first sample, than wash and run the second one (the second bacteria)?

You can monitor the sequencing run in real-time and stop it once the desired coverage is reached.

I have seen people run 12 bacteria on 1 minion flowcell with multiplexing. This worked very well, but they did have high quality DNA and experience and got >20GB of data from the flowcell (not too easy AFAIK). But I think the multiplexing approach is better than the subsequent sequencing approach.

Keep in mind most data is generated at the start of the minion run, so 0.5h-1 hour of sequencing might be sufficient for a 3 MB bacterial genome. But this is different for every sample/flowcell/run. Make sure your HMW dna is good and freshly extracted, this is critical to get the long reads you need to ease assembly.
