What is the difference between vst and rlog to perform PCA ? and must I make vst or rlog before PCA for RNA sequencing data showing differentially expressed genes?

When I wrote t-test to check if the cancer samples are significantly different than the normal samples using R I was thinking whether I should add a sentence encouraging you to google your problems first but I did not. Now I do. All these standard questions have been asked before. rlog and vst are even covered in the DESeq2 manual and this has been asked many times before. You have asked many questions here over the last two years. Nothing basically wrong with that, but an essential skill to build a career in bioinformatics relies on finding solutions and answers without spoonfeeding from others. Use search engines, read manuals and papers, do research yourself. You cannot expect strangers on the internet to guide you through your analysis. There are a few users here who assume biostars was a personal helpdesk . Please build the essential skills to find answers yourself, especially if questions have been asked before, don't become one of these lazy users who shotgun questions. Pretty much all relevant questions related to the strategy in the other thread have been asked before, please go find the answers, that is also a learning experience. There is in the longterm no value in getting spoon-fed.

I really don't want to waste your time or my time to defend or to say that I tried on internet and I opened many tutorials and Youtube videos but I can't find a direct or clear answer or an answer that fulfill my curiosity. In addition, regarding your answer to my previous question, I did all what you said before I post the question but you really don't know how is the data that I have because it's not real so I can't perform GSEA, DAVID analysis etc.

Try to be kind with people either answer either ignore the question and don't blame yourself for helping others and don't mention a favor you have done because it's not a good way and does not reflect something good of you, better not to help then. it's good to advise people but remember that you only see posts but you don't see the effort that has been done before posting the questions. In addition, for the posts that I have asked before for the last years that you mentioned I also put an answer when I find so I also help others and others helped me as it's normal in research to help and to be helped. That is not what the job description of a moderator is, simple as that. Users who do this spoon-feeding behaviour often get abandoned by the users who can provide quality answers, so this is not what you want. It's an advise, you can ignore it if you want of course.
