scRNA-seq analysis after overexpressing a specific gene

Hello all,

I performed scRNA-seq analysis after overexpressing a specific gene in a particular tissue and isolating the tissue. However, upon reviewing the sequencing results, I found that the gene in the overexpression group did not show a significant increase. Moreover, transcripts of the gene were not highly captured at all.

Could this issue stem from the technical limitations of scRNA-seq? (I used the Chromium kit from 10x Genomics.)

Unfortunately, I cannot perform additional experiments, such as qPCR, as all tissue samples were used for the scRNA-seq analysis. In this case, is it still possible to derive meaningful analytical results even if the overexpression of the gene is not significantly evident in the scRNA-seq data?

What was the method of overexpression? Is the OE gene the same species as your tissue?

Do you have any evidence the OE worked?

I think it's possible it's a detection issue, but sort of seems unlikely to me in an OE scenario
