Hello there. I am doing a research on tick cell culture of IDE-8 cell. But I am confused about the presence of cell at 40x magnification. my cell types are suspension type and literature shows it is adherent. And i found no cell in cytocentrifuge slide smear. I want to do bioinfor.atics analysis after my cell culture is done. But I am not sure about the cell. Can you suggest me any other way?

Ultimately, if an adherent cells is in suspension, that is due to a lack of attachment. This could be the substrate, or type of material the cell is grown on. Other considerations would be the health of the cell. If you saw them in culture but then no cells after collecting them and doing a cytospin, then I would question your collection technique and then if the cells were dense enough when loaded in the cyto-centrifuge. Another consideration is if the funnel made good contact with the slide, if there's a leak, then all the cells would escape.
It sounds like there may be multiple issues, so the best option here would be to try and find someone to physically show you how to do it. You may also try to find some videos or something and double check your work.
