Given a protein sequence, is it there any experimental method that allows to detect a given triplet and change or mutate it to another one?

If by "experimental method" you mean "in the laboratory", then this is not a bioinformatics question and should be closed. And if you're looking for publications, you should learn how to search literature effectively.

Triplet is a confusing word. Are you asking about a codon triplet, or, as I interpret your questions, do you want to know about detecting a 3-amino acid sequence within the larger polypeptide? Both can be detected and can be mutated.

I think you can do it by site directed mutagenesis
