I am interested in assembling the genome of a micro-parasite, for which the DNA I can obtain to amplify is on the order of picograms. I read a paper (Lasken 2009, Genomic DNA amplification by MDA method) that advocates use of Multiple Displacement Amplification using phi29 polymerase to amplify very small amounts of DNA. Among other things in the paper, it is mentioned that MDA performs well for samples that would be later sequenced using dideoxy approach and can produce upto 4000x amplification. I am interested in knowing opinions of experts regarding this before I settle on using MDA for my work. Any inputs would be welcome.
