Building Human Gene Annotation File from Gene Primers

Hello, I am trying build human gene annotation file (either GTF or GFF2) works from gene primers, which could be used for processing raw rna-seq file. So, I have some genes forward and reverse primers such as gene LIF forward primer TACCGCAT, now using it I am trying to human gene annotation file. This file will later be used for alignment/assembly purpose. The goal of this is to map qPCR data to RNA-seq data. Can anyone please help me with this with the steps? Thanks
