Generate_FASTQ Module on NextSeq: Manifest?

Hello! I need to obtain FASTQ files for a WES Project using a NextSeq 550. If I use the Generate FASTQs Module, I do not need to load a Manifest, korrect?

If by manifest you mean a samplesheet that describes samples in the pool that is used for demultiplexing then you will need to include it. But if the manifest includes exome regions then it may be needed if you are going to use onboard DRAGEN for analysis.

The Manifest has the positions of the enriched regions. In the Sample sheet I can give the name of the Manifest - if needed. In another file there are the names and sequences of the indices for demultiplexing (Library Kit file).

I load the Manifest file using the Module DNA Enrichment, but I am not sure if I need it with the Module Generate FASTQs.

I assume you are referring to on board DRAGEN for demultiuplexing and analysis. For generating fastq you should not need the manifest.
