Hello I have knocked down the LEMD2 gene in the HLE-B3 cell line using siRNA. Also, I set up a control using nontargeting siRNA. On doing RNAseq and differential gene expression analysis, not many genes got differentially expressed (just n=10 were DEG at p<0.05). These are the genes after adjusting for benjamin hochberg test. what could this tell about the LEMD2. or what could be wrong as I am not seeing many genes being DEG. Thanks

give more about experimental details like how many replicates used for the same so that other can give their insights

I have used three replicates for LEMD2 siRNA treated cells and 3 replicates for Non targeting siRNA treated cells. Cells were recovered after 72 hrs and knock down efficiency was checked using qRT-PCR and western blotting. qRT-PCR showed ~85% downregulation of LEMD2. Role of LEMD2: This gene encodes a LEM domain-containing transmembrane protein of the inner nuclear membrane. The protein is involved in nuclear structure organization and plays a role in cell signaling and differentiation. Mutations in this gene result in Cataract 46, juvenile-onset. Multiple transcript variants have been found for this gene.
