Hello.

I have been trying to do post-sequencing analysis (i.e., basecalling) in MinKNOW using the raw reads I generated from my sequencing run. However, it always gives me this error:

2025-01-28 14:23:04.442677 [ont/error] boost::filesystem::directory_iterator::construct: Operation not permitted [system:1]: "/Users/cedrickalguzar/Downloads/queued_reads/complete_reads_f025be2b-1b99-428a-82eb-02f6839905ce"
2025-01-28 14:23:04.442701 [ont/warning] An error has occurred. Aborting.

Does anyone here know how to troubleshoot this? Thank you!

Since you added the dorado tag are you directly using dorado to recall bases or are you using the MinKNOW GUI? You will need POD5/fast5 files to recall bases using a different accuracy model. Do you have those available?

If that is not applicable you may want to contact ONT support and see if they can help.

I am using MinKNOW GUI for my post-sequencing basecalling and the data that I have been trying to load are all POD5 files.
