Hi

I am very new to bioinformatics and I am reading this paper but I have no Idea what it is talking about :)

Can anyone help me, or at least tell me what prerequisites I need for reading this paper and other papers in this field?

Regards
Hadi

Start with Wikipedia and understand each term like Next Generation Sequencing, RNA-Sequencing, genes, transcripts etc.

I was browsing books in bioinformatics and honestly the didn't really help me, they were all basic. which chapters in text books do you suggest?

Hi,

Transcript expression level, refers to how abundant a transcript is, i.e. if a certain mRNA (a transcript) exist in many or few copies (expression level).

DE analysis is the analysis of Differentially Expressed genes. It implies comparing relative abundance of mRNAs between different conditions and thereby identifying genes that are differentially expressed (transcribed more or less in one condition compared to the other).

I would recommend reading this paper: http://www.ncbi.nlm.nih.gov/pubmed/26248053

Good luck
