I have a gene with 120075 bp and 7 exons that include 1961 bp but its gene expression is 543 aa. why it isnt exactly (1961/3) = 653 ?

In addition to alternative splicing as suggested by natasha.sernova you also have a 5' and 3' UTR: untranslated sequences containing mainly regulatory fragments which are therefore not translated. Depending on the organism you are working on you will find plenty of information on sequence annotation, e.g. in the UCSC genome browser or ensembl genome browser.
I would suspect alternative splicing.
