I just want to call the attention to a remarkably detailed comment to a question that seems to be of great interest to many (btw make sure to vote for those posts and not this one):

Hi Istvan,

I found this when I was, um, self-googling...

I've made some updates to it. There is some nice work in yeast with some highly replicates samples now that may superscede some of it , but I think most of it is still relevant.
