What Pacbio HiFi data do people save ?

Hello all,

what PacBio data is worth saving long term?

  • fastq
  • bam (does this have additional signal for methylation calls etc )
  • others ?

Ok, to answer my own question, bam is required for methylation analysis with the pacbio primrose software, so might be worth keeping too.

If you have the means/budget then saving a copy of all raw data a sequencer produces (for some period of time) is likely a good policy, especially if you are a core and expect clients to ask for additional data. I assume you release fastq sequence as a minimal deliverable?

That's good advice, if affordable. I just have little practical experience with Pacbio Hifi yet. In my core days we kept all data, even if there was never once a request for alternative demux from the bcl files.
