How to create Illumina sample sheet file for single index data?

Hi All

I need help regarding how to format the sample sheet file (required for demultiplexing using bcl2fastq) for single index data. Usually I have dual index data and I keep below column names in the sample sheet.

  • Sample_ID

  • Sample_Name

  • Sample_Plate

  • Sample_Well

  • I7_Index_ID

  • index

  • I5_Index_ID

  • index2

  • Sample_Project

  • Description


Now, this time I have single index data and I am wondering whether I need to -

[1] Keep the i5_index_ID & index2 columns but keep them blank 1

OR

[2] Keep the i5_index_ID & index2 columns and add some text (NA) 2

OR

[3] Delete i5_index_ID & index2 columns 3

Please help.

Remove the I5_Index_ID and index2 columns. BTW, you can always remove Sample_Plate and Sample_Well.

You can download Illumina experiment manager software (Windows only). It allows you to make correct samplesheets for all Illumina kits/sequencers.
